The story begins in 1967, when a pregnant woman is attacked by a vampire in the delivery room. Bitten on the neck, she gives birth to a son and then dies, but the infant survives with vampire blood mixed into his own—this child is the one who will later become Blade. Raised and trained from a young age by the vampire hunter Whistler (played by Kris Kristofferson), Blade possesses the superhuman strength and endurance of a vampire while being immune to their weaknesses (able to operate during the day), becoming a "Daywalker" who walks between the worlds of humans and vampires.

Blade is dedicated to eradicating vampires, wielding custom weapons such as silver-edged swords and flame throwers as he hunts alone in the darkness. Meanwhile, the ambitious vampire leader Deacon Frost (played by Stephen Dorff) seeks to awaken the ancient blood god and unleash a "blood tide" that would turn all of humanity into vampires. To achieve this, Frost not only confronts Blade directly but also attempts to use Blade's half-human, half-vampire lineage for a ritual sacrifice. With the help of hematologist Karen Jenson (played by N'Bushe Wright), Blade must stop Frost's conspiracy and come to terms with the truth about his own origins.
